Abstract [eng] The data of reaction kinetic studies of glucose oxidation, catalyzed by gold nanoparticles (AuNPs), glucose oxidase (GOx) or AuNPs and GOx conjugates, is submitted in the final thesis for the master's degree. The influence of AuNPs size on the kinetics of glucose oxidation reaction was investigated by chronoamperometric analysis using an analytical potentiostat system with a Clark's oxygen electrode. The experiment was carried out using different substrate – glucose – concentrations and AuNPs varying in diameter. During the analysis, the change of oxygen concentration was recorded, from which the initial reaction rate was calculated. Examining the differences, it was found that the initial reaction rate increases when AuNPs diameter increases from 13 nm to 50 nm. Also, immobilization of GOx on AuNPs surface has no significant effect on the kinetics of the enzyme, but increasing immobilized GOx concentration on the surface of AuNPs lowers the initial rate of the AuNPs catalyzed glucose oxidation reaction. The work consists of six parts: introduction, review of scientific literature, methods and tools, results and their discussion, conclusions and literature. Thesis consists of: 49 pages, 17 figures, 4 tables and 56 bibliographical entries.
